
The Incucyte®️ Cell Cycle Green/Red Lentivirus Reagent is a fluorescent, single cassette indicator expressing both the GFP (green fluorescent protein) and mKate2 (red fluorescent protein) to distinguish between cells in the G1 and S/G2/M cell cycle phase without altering cell function. This reagent is ideal for generating stable cell populations or clones using puromycin selection. The Incucyte Cell Cycle Green/Red Lentivirus Reagent has been validated for use with the Incucyte® Live-Cell Analysis System for the real-time, integrated analysis pf spatial and temporal patterns of cell cycle dynamics. Cell cycle analysis can be multiplexed with label-free cell counts using the Incucyte® Cell-by-Cell Analysis Software Module for quantification of the total cell population to determine percentage of cells within each cell cycle phase.
Visualizing synchronized division of HeLa cells stably expressing Incucyte® Cell Cycle Green/Red Lentivirus Reagent. Division of HeLa cells stably expressing Incucyte® Cell Cycle Green/Red Lentivirus Reagent was arrested by treatment with Thymidine (2.5 mM, 0 - 24h), causing cells to accumulate in G2/M (green). Upon removal of the Thymidine block, cells begin to synchronously divide and progress through the cell cycle (24 – 72h). Data shows the % cells expressing each marker peaks in sequence: non-fluorescent (M – G1 transition), red (G1), red + green (G1 – S transition), green (G2/M). Two full cycles of cell division are observed for this cell type between 24 and 72h.
